The clustered regularly interspaced short palindromic repeats (CRISPR) system has revolutionized the genome engineering field thanks to its versatility, ease of use and robustness. Emerged as an adaptive immune system in bacteria, further understating and engineering of the CRISPR/Cas system has opened to possibility to use this technology for precise manipulation of genetic information in cells. The early successes have initiated an explosive growth in the discovery and development of additional Cas-based systems, which nowadays constitute powerful tools driving major advances in the life sciences. Here, we will discuss the large plethora of Cas9 technologies and their applications from fundamental science to translational medicine, providing an overview of the capabilities as well as of the current limitations of CRISPR technologies in a way that facilitates tool selection.
