Abstract
For cells to function properly, it is necessary for cell organelles to communicate with each other. In this context, mitochondria establish functionally crucial connections with virtually all other intracellular structures. Specifically, mitochondria primarily interact physically with the Endoplasmic Reticulum (ER) and the membrane surface of lipid droplets (LDs). These interactions not only impact mitochondrial behavior and activity but also influence a wide variety of cellular functions.
Here, we provide two examples illustrating the importance of these mitochondria-organelle contact sites in different physio-pathological conditions. Firstly, we describe how the association between ER and mitochondria affects the process of wound healing by regulating the activation of a specific inflammatory pathway. Secondly, we demonstrate that a specific dysregulation of the mitochondrial Ca2+ machinery promotes the formation of aberrant LD-mitochondria contacts, which in turn characterize the progression of clear cell renal cell carcinoma.
