Abstract
A fundamental question in the field of neurodegeneration is why certain brain regions and cell subpopulations are selectively vulnerable to degeneration. The most prominent example is Alzheimer's disease (AD). From the early stages of this disease, only certain neurons accumulate deposits of tau protein, which colocalizes with heparan sulfates. Heparan sulfates are produced by a biosynthetic machinery involving dozens of genes, some of which show specific tissue and cell subtype enriched expression, suggesting that these genes may represent susceptibility factors involved in the selective vulnerability of cells and brain regions to degenerate in AD. Here, we used spatial transcriptomics and RNAseq technologies in combination with histological, biochemical, cellular, and in vivo approaches to demonstrate that heparan sulfate biosynthetic enzymes represent vulnerability factors expressed in brain regions and neuron subpopulations that selectively degenerate in AD.
